SONIC: Subcallosal Cingulate DBS for Treatment-Resistant Depression (NCT03437928)

ClinicalTrials.gov4/1/2026ยท04/01/26๐ŸŒ USA

Summary

Multicenter trial of subcallosal cingulate white matter DBS in 60 severely treatment-resistant depression patients across 6 US sites, using diffusion tractography-guided targeting to identify the optimal stimulation site per individual. Extends Emory's pioneering SCC DBS work with a standardized tractography-based surgical protocol.

Why it matters

SCC DBS has shown remarkable antidepressant effects in open-label series but mixed pivotal trial results. Tractography-based precision targeting may resolve the inconsistency if targeting variability was driving previous negative results.
